Dosage

Daraprim is available in the strength of 25 mg tablets only.

Usage

Daraprim (Pyrimethamine) is a prescription medication used with a sulfonamide to treat a serious parasite infection (toxoplasmosis) of the body, brain, or eye and to prevent toxoplasmosis in people with HIV infection. Daraprim is also used with chloroquine to treat people with acute malaria. The active ingredient in Daraprim is Pyrimethamine. Pyrimethamine belongs to a class of drugs known as antiparasitic, an antimalaria agent. It works by killing parasites. Pyrimethamine is well absorbed, with peak levels between 2 to 6 hours after swallowing the tablet. After that, it is eliminated slowly and has a plasma half-life of approximately 96 hours.

Side Effects and Precautions

The common side effects of Daraprim are nausea, vomiting, abdominal pain, abnormal skin pigmentation, lightheadedness, and insomnia.

Stop using Daraprim and contact your doctor right away if you have any of the serious side effects such as:
• Severe cramps;
• Severe diarrhea

Warnings and Interactions

Before taking Pyrimethamine, tell your doctor or pharmacist if you are allergic to it; or have any other allergies. Daraprim is a tablet and should be taken by mouth, usually once or twice daily or according to your prescription. Take this medication with food to reduce the risk of nausea and vomiting. If you are vomiting continuously, your doctor may lower your dose or direct you to stop taking this medication. It is often recommended to use Daraprim with folic acid to prevent blood problems caused by Pyrimethamine. It would be best to follow your doctor's directions carefully. If you take a "sulfa" medication with Pyrimethamine, drink enough water to avoid kidney problems. This medication works best when the amount of drug in your body is kept at a constant level. The duration you will use the Daraprim tablet depends on the nature of your sickness and response to treatment. You should not increase your dose without your doctor's approval or skip your medication. Skipping or changing your dose without consent from your doctor may cause the number of parasites to increase, make the infection more difficult to treat (resistant), or worsen side effects. Tell your doctor if your condition lasts or gets worse. Before you start using Daraprim, inform your doctor about your medical history, especially if you have the following:
• Anemia, particularly megaloblastic anemia;
• A blood disorder such as G6PD deficiency;
• Kidney problems;
• Liver problems;
• Seizure

Tell your doctor about all your medications, including prescriptions, over-the-counter medications, herbal products, and vitamin supplements. Tell your doctor if you are pregnant or plan to become pregnant. During pregnancy, this medication should be used only when needed. Discuss the risks and benefits with your doctor. Folic acid is essential during pregnancy. Your doctor will prescribe folic acid to prevent low folate levels. It would be best to know that Daraprim passes into breast milk. Consult your doctor before breastfeeding. There may be a drug interaction between Daraprim and other drugs such as eliglustat, trimethoprim, zidovudine, methotrexate, daunorubicin, and proguanil.
