Usage

Gavreto (Pralsetinib) is a prescription medication used to treat advanced stages of non-small cell lung cancer (NSCLC) in adults. Gavreto is also used as an alternative medicine for treating adults and children 12 years of age and older with advanced thyroid cancer or metastasized thyroid cancer and those that have used radioactive iodine with no significant improvement. This drug is only prescribed if the lung cancer is present with a specific rearrangement in a gene called rearranged during transfection (RET). The active substance in Gavreto is Pralsetinib. Pralsetinib belongs to a class of medication called RET inhibitors. It works by slowing or stopping the growth of cancer cells.

Side Effects and Precautions

The common side effects of Gavreto are constipation, edema, muscle pain, cough, and tiredness.

Stop using Gavreto and contact your doctor right away if you have any of the severe side effects such as:
• Fever and chills;
• Signs of an allergic reaction such as breathing difficulties, hives, swelling of the face, lips, or throat;
• Unusual bleeding;
• Worsening cough;
• Wounds that refuse to heal

Warnings and Interactions

Before using Pralsetinib, tell your doctor or pharmacist if you are allergic to it or have any other allergies. Gavreto comes as a capsule. The capsule should be taken once daily. Swallow the capsule whole and do not crush, chew, break, or open it. Take this drug with a glass of water on an empty stomach. Do not eat for at least two hours before and at least one hour after taking this drug. If you vomit after taking Pralsetinib, do not take another dose. Continue with your regular dosing schedule. Your blood pressure will need to be checked often, and you will need frequent blood tests while taking this drug. Avoid driving or using machines while using this drug. Gavreto can make you tired. Do not consume grapefruit products or Seville oranges while using this drug. Avoid alcohol when using this drug. Drinking alcohol can increase the side effects of the drug. Discuss with your doctor if you experience a serious side effect of this medication.

Before you start using Gavreto, inform your doctor about your medical history, especially if you have:
• Bleeding disorders;
• High blood pressure;
• Kidney disease;
• Liver problem;
• Other lung or breathing problems besides lung cancer;
• Plan to have surgery;
• Tuberculosis

Tell your doctor about all your medications, including prescriptions, over-the-counter medications, herbal products, and vitamin supplements. Tell your doctor if you are pregnant or plan to become pregnant. Gavreto may harm an unborn baby. Use non-hormonal birth control while using this medicine and for at least two weeks after your last dose. Gavreto may make hormonal birth control less effective. Males with female partners of childbearing potential should use contraception during treatment and for 1 week after completing the last dose. Tell your doctor right away if pregnancy occurs. It is not known if Pralsetinib passes into breast milk or if it may harm a nursing infant. Do not breastfeed while using this drug and for at least one week after your last dose. The use of Gavreto should be avoided in children less than 12 years old. There may be a drug interaction between Gavreto and other drugs, such as ketoconazole, phenytoin, phenobarbital, St. John’s wort, nefazodone, rifampicin, etc.
