Usage

Giotrif (Afatinib dimaleate) is a prescription medication used to treat metastatic non-small cell lung cancer (NSCLC) in patients whose tumors have activating epidermal growth factor receptor (EGFR) mutations. NSCLC hardly shows symptoms until the disease has well advanced. However, early detection of the symptoms can help. Afatinib dimaleate is also used to treat locally advanced or metastatic NSCLC of squamous histology progressing on and after platinum-based chemotherapy. The active ingredient in Giotrif is Afatinib dimaleate. Afatinib dimaleate belongs to the class of medication called protein kinase inhibitors. It works by blocking the signals (messages) of epidermal growth factor receptors (EGFR) that tell cancer to divide and grow. This helps to slow down or stop the cancer from growing. People with EGFR-negative tumors should not use Afatinib dimaleate.

Side Effects and Precautions

The common side effects of Giotrif are appetite loss, diarrhea, heartburn, runny nose, and weight loss.

Stop using Giotrif and contact your doctor immediately if you have any of the following serious side effects such as:
• Eye inflammation;
• Severe skin peeling or blistering;
• Signs of a serious allergic reaction, such as breathing difficulties, skin rashes, stomach upset, swelling of the face, mouth, tongue, or throat;
• Signs of low potassium levels in the blood, such as weakness, fatigue, irregular heartbeat

Warnings and Interactions

Before taking Afatinib dimaleate, tell your doctor or pharmacist if you are allergic to other protein kinase inhibitors or have any other allergies. Giotrif is a tablet, and it should be taken orally (by mouth) as directed by your doctor, usually once daily. Follow your prescription label carefully. Swallow the tablets whole with water. You may dissolve the tablet in non-carbonated drinking water if you have swallowing difficulties. Do not use other liquids. Drop the tablet in half a glass of drinking water and stir occasionally for up to 15 minutes until the tablet is broken into very small pieces. Drink the liquid immediately. Rinse the empty glass with half a glass of drinking water and drink it. Food can interfere with the absorption of Giotrif. Take Afatinib dimaleate at least 3 hours after eating or 1 hour before eating. If you vomit shortly after taking a dose, do not take another dose. Continue with your regular dosing schedule. You may have diarrhea while using this medication. Your doctor may prescribe an anti-diarrhea medicine like loperamide when starting treatment with Giotrif. Drink plenty of fluids and take anti-diarrhea medicine if you notice signs of diarrhea. Your doctor may adjust your dose depending on how well you tolerate Afatinib dimaleate. Do not stop taking this medication without consulting your doctor. Your condition may become worse when this medication is suddenly stopped. Giotrif may cause your skin to be much more sensitive to sunlight than it normally is. Limit your exposure to sunlight while you are taking Giotrif. When you are outdoors, wear a hat, protective clothing, and sunscreen.

Before you start using Giotrif, inform your doctor about your medical history, especially if you:
• Are lactose intolerant;
• Become pregnant or nursing a child;
• Have gastrointestinal problems;
• Have had cancer that has spread to the bowel;
• Have heart problems;
• Have interstitial lung disease;
• Have kidney problems;
• Have liver problems

Inform your doctor about all your medications, including prescriptions, over-the-counter medications, herbal products, and vitamin supplements. Tell your doctor if you are pregnant or plan to become pregnant. Using Giotrif in pregnancy can harm your unborn baby. Use effective birth control when taking this drug and for at least two weeks after your last dose. Afatinib dimaleate may pass into breast milk and affect your baby. It would be best to know that breastfeeding is not recommended while using this drug. Consult your doctor before breastfeeding. There may be a drug interaction between Afatinib dimaleate and other drugs such as aubagio, Nexavar, venclexta, and Betaseron.
