Mechanism of Action
Gefitinib selectively and reversibly inhibits the intracellular tyrosine kinase domain of the EGFR (epidermal growth factor receptor), also known as ErbB1. In lung cancers with activating EGFR mutations, the receptor is constitutively active, continuously driving cancer cell growth, survival, and spread. By blocking EGFR kinase activity, gefitinib disrupts these cancer-promoting signals, leading to cancer cell growth arrest and cell death. Because it is selective for the mutated EGFR found in cancer cells, gefitinib can be effective while causing less harm to normal cells compared to conventional chemotherapy.
