Mechanism of Action
Trametinib is a selective, allosteric inhibitor of MEK1 and MEK2 (mitogen-activated protein kinase kinases), which are downstream effectors in the BRAF-MEK-ERK signaling pathway (also known as the MAPK/RAS-RAF-MEK pathway). In cancers with BRAF V600 mutations (such as V600E or V600K), the BRAF kinase is constitutively active, continuously activating MEK and driving cancer cell proliferation, survival, and spread. By blocking MEK1 and MEK2, trametinib disrupts this signal and stops tumor cell growth. When combined with a BRAF inhibitor (dabrafenib), which directly blocks the upstream mutated BRAF protein, the combination provides a more complete and durable blockade of the pathway, reducing the likelihood of resistance compared with either drug alone.
